Eligibility Advanced Stem Cell Techniques is an advanced hands-on program open to residential and international students of all levels – undergraduate and postgraduate.  Applicants must have completed at least 1 year of an undergraduate degree in a bioscience related subject covering advanced cell biology.  To attend this course, you must be proficient in aseptic technique and have prior experience culturing mammalian cells in a professional cell culture setting. Students who do not have cell culture experience can gain the required skills by attending our essential cell culture techniques program.

Description

“Advanced Stem Cell Techniques” provides comprehensive training in validated methods for deriving, culturing, characterizing, and differentiating three types of human stem cells – neural (NSC), mesenchymal (MSC) and induced pluripotent (iPSC).

Curriculum

Participants learn to:
– Derive NSC from hiPSC
– Culture NSC as adherent monolayers and neurospheres
– Characterize and differentiate NSC
– Culture, characterize and differentiate MSC
– Perform cloning assays
– Reprogram hiPSC using Episomal plasmids and Sendi Virus
– Select, prepare and aliquot hiPSC/hESC culture reagents
– Morphologically assess hiPSC/hESC colonies
– Culture hiPSC/hESC under feeder free conditions in mTeSR and NutriStem
– Pick and manually passage hiPSC/hESC colonies
– Passage hiPSC/hESC using Accutase and ReLeSR
– Cryopreserve and thaw hiPSC/hESC
– Prepare embryoid bodies (scraped and spin-EB’s)
– Differentiate hiPSC/hESC to specific germ layers
– Electroporate hiPSC/hESC
– Establish an hiPSC/hESC laboratory
– Perform standard characterization for immunological markers, vector integration and pluripotency
– Quality control monitor hiPSC/hESC during prolonged culture
– Electroporate hiPSC/hESC
– Troubleshoot common problems
– Comply with ESCRO regulations
– Reduce costs through optimized purchasing and laboratory organization

Our streamlined hiPSC protocols have been validated for use with human embryonic stem cells (hESC). The course curriculum is updated annually to reflect current techniques.
